Nah Paul Dino who worked on Batman TAS invented her for the show as a cameo for his friend who voiced her.
At the time Arleen Sorkin was famous so the joke was that Joker was being taken care of someone who was famous.
But Paul expanded the idea to her being Jokers girlfriend because people liked the chemistry.
Harley is basically a cameo parody of a famous actress that became more famous then the actress.
